چکیده
Dermatomyositis (DM) is a chronic, systemic autoimmune disease affecting the skin, muscle, and lungs. Lenabasum, non-psychoactive cannabinoid type 2 receptor (CB2R) agonist, currently being investigated as non-immunosuppressive treatment option for DM. The activation of CB2R has been shown to reduce several key pro-inflammatory cytokines implicated in Our lab previously demonstrated via immunohistochemistry (IHC) that Lenabasum decreases CD4, IFNβ, IFNγ IL31 expression DM skin at 12 weeks (p<0.05), with no differences IL4 compared placebo (p>0.05). In this study, we then utilized multiplexed flow cytometry leukocytes eluted from further analyze on cell lineages. When evaluating lineages, CD4 T helper (Th) subsets were gated CD4+ IFNγ+ Th1 CD4+IL4+ Th2. There was significantly higher frequency parent (FOP) percentage (p<0.05) (61.05%, n=6) versus Th2 cells (25.5%, n=6). Among myeloid there greater FOP M2 macrophages (CD68+CD163+), CD14++CD16++ macrophages, monocyte derived dendritic (moDCs; CD11c+CD14+). With 15mmol trend towards decrease of: TNFα macrophages; IFNβ Tcells; CD16+ cells; cells, CD14++CD16+ macrophages. also significant moDCs secreting (p<0.01). Imaging mass untreated highest MPI moDCs. elevated lesser extent other These data suggest may exert specific anti-inflammatory effects DM, particularly Th1-derived IL31.

Anti-nociceptive interactions between opioids and a cannabinoid receptor 2 agonist in inflammatory pain
The cannabinoid 1 receptor and cannabinoid 2 receptor can both be targeted in the treatment of pain; yet, they have some important differences. Cannabinoid 1 receptor is expressed at high levels in the central nervous system, whereas cannabinoid 2 receptor is found predominantly, although not exclusively, outside the central nervous system. The objective of this study was to investigate potenti...
متن کاملChanges in motor and learning behaviors of rats prenatally exposed to WIN 55212-2, a cannabinoid receptor agonist
Abstract* Introduction: Cannabis consumption during pregnancy may affect fetal growth, motor performance, memory and cognitive functions. Methods: Primiparous pregnant Wistar rats were randomly assigned to 4 treatment groups (6 rats per group) consisting of control and sham groups as well as 2 groups treated with Win. Win treated groups received daily s.c. injections of 0.5 or 1 mg/kg WIN ...
متن کاملRole of Nitric Oxide in the Antipruritic Effect of WIN 55,212-2, a Cannabinoid Agonist
Introduction: For centuries, cannabinoids are known to be effective in pain relief. Itch is an unpleasant sensation that provokes a desire to scratch. Since itch and pain are two sensations sharing a lot in common, we aimed to investigate whether the cannabinoid agonist WIN 55,212-2 reduces serotonin-induced scratching behavior and also observe whether modulation of nitric oxide (NO) production...
متن کاملNanoliposomal Nitroglycerin Exerts Potent Anti-Inflammatory Effects.
Nitroglycerin (NTG) markedly enhances nitric oxide (NO) bioavailability. However, its ability to mimic the anti-inflammatory properties of NO remains unknown. Here, we examined whether NTG can suppress endothelial cell (EC) activation during inflammation and developed NTG nanoformulation to simultaneously amplify its anti-inflammatory effects and ameliorate adverse effects associated with high-...
متن کاملEffect of Cannabinoid Receptor Agonist WIN55, 212-2 on the Anxiety Induced by PTSD in Male Rats
Introduction: Posttraumatic stress disorder is a severe anxiety disorder caused by exposure to traumatic events. The aim of this study was to induce PTSD in rats and examine the effect of WIN55-212-2, a cannabinoid receptor agonist, on anxiety. Methods: SPS&S model was used to induce PTSD in 56 male Wistar rats. Rats were restrained for 2 h, immediately followed by forced swimming for 20 mi...
